Dulbecco's Modified Eagle Medium (DMEM) is a widely used basal medium. It was originally modified by Renato Dulbecco based on Eagle’s Minimum Essential Medium (EMEM) to support the in vitro growth of mammalian cells, especially adherent cells.
DMEM - Low Glucose is the low - glucose version of DMEM, with a glucose concentration of 1.0 g/L (5.5 mM), which is closer to the in vivo blood glucose concentration. It is suitable for the culture of cells that are sensitive to glucose concentration or tend to be in a normal metabolic state. What distinguishes DMEM from other media is that it contains amino acids and vitamins at a concentration four times that of the original Eagle's Minimum Essential Medium. DMEM does not contain proteins, lipids, or growth factors. Therefore, DMEM needs to be supplemented with nutrients, and it is usually necessary to add 10% fetal bovine serum (FBS). DMEM uses a sodium bicarbonate buffering system (3.7 g/L), so an environment with 5–10% CO₂ is required to maintain the physiological pH value.
